Abstract
For the determination of isomaltooligosaccharides in foods such as dairy products, solid drinking, candy and healthy food, a high performance liquid chromatography method was established. Isomaltose (IG2), panose (P), isomaltotriose (IG3) and isomaltotetraose (IG4) in various foods were separated on a NH2 column by gradient elution and detected by evaporative light-scattering detector (ELSD). The linear ranges of the calibration curves for IG2, P, IG3 and IG4 were 0.1~0.45mg/mL, 0.1~0.45mg/mL, 0.2~0.65mg/mL and 0.1~0.35mg/mL, respectively. The linear correlation coefficients (R2) were 1, 0.9999, 0.9998 and 0.9999, respectively. As to different kinds of foods, the spike recoveries were in the range of 98.0%~105.0%. Coefficient variability of repeatability and reproducibility were less than 10%, respectively, except the reproducibility of isomaltotetraose. The method was simple and was apt to be grasped, can be used in the quantification of isomaltooligosaccharides in complex food matrix.
