Abstract
【Objective】 To study the effect of Fasudil hydrochloride(Fasudil) preventing acute renal injury in rats caused by rhabdomyolysis and explore its action mechanism.【Methods】 Forty-five male SD rats were randomly divided into three groups: control group,acute renal failure(ARF) group and treatment group.ARF group were injected with 50% glycerol.Blood samples were taken from every rats to detect the levels of cretonne(CRE) and urea nitrogen(BUN).TUNEL analysis was used to determine the apoptotic death.The cleaved caspase-3 was detected by western blot analysis.【Results】 The blood levels of CRE and BUN in ARF and treatment group was higher than that in control group(P 0.01).However,The CRE and BUN levels in treatment group were lower than that in ARF group(P 0.01).The percent of apoptosis in control,ARF,and treatment group were(2.14±1.66)%,(56.94±11.35)%,and(26.17±7.82)%,respectively.Compared with ARF group,treatment group has a lower apoptotic death.The cleaved caspase-3 was detected in ARF and treatment group by western blot analysis.The expression of cleaved caspase-3 could be inhibited by Fasudil treatment.【Conclusion】 Fasudil inhibited the apoptosis to protect renal against rhabdomyolysis-induced acute renal injury.
