Abstract
OBJECTIVE To study the synergism of bactericidal activities of fleroxacin (FLX) combined with azithromycin(AZT) in the treatment of local infection caused by Pseudomonas aeruginosa biofilm. METHODS The tissue cage method was used to establish the animal model of local P. aeruginosa biofilm infection and this model was confirmed by using microbiological method, histological method and scanning electronic microscopy (SEM). The synergism of AZT combined with FLX was studied by using microbiologic method and SEM. RESULTS When tissue cages with P. aeruginosa biofilms were implanted into subcutaneous tissue of rats, biofilms could develop in the surface of tissue cages in vivo and caused local infections, which could be confirmed by histological, cytologic and mirobiologic methods as well as SEM. When 75 mg/kg AZT was administered once per day the concentration of AZT in tissue cage fluid was above its 1/16 MIC to P. aeruginosa. When 25 mg/kg FLX were administered twice per day its concentration in tissue cage fluid was above its MIC to P. aeruginosa. When FLX was administered with AZT viable counts in tissue cage fluids were less than that when FLX was administered alone. Bacteria on the surface of tissue cage were also less as confirmed by SEM. CONCLUSIONS Azt could strenghthen the efficacy of FLX in the treatment of local biofilm infections.
