Abstract
Ethyl 7-fluoro-4-oxo-1,4-dihydroquinoline-2-carboxylate was prepared from sodium salt of diethyl 2-oxosuccinate and 3-fluoroaniline via nucleophilic addition reaction and cyclization.The effects of the proportion of raw materials,reaction temperature and reaction time on the yield of the product were analyzed,and optimum process conditions were determined.The first step reached the highest yield at temperature 55 ℃,reaction time 15 h and n(sodium salt of diethyl 2-oxosuccinate)∶n(3-fluoroaniline)=1∶1.6,and its yield reached 54.52 %.The second step reached the highest yield at temperature 265 ℃,reaction time 15 min,and its yield reached 79.88 %.The total yield was 43.55 %.The structure of target product was confirmed by MS and 1H-NMR.
