Abstract
Carbobenzoxy-L-leucyl-L-valyl-S-benzyl-L-cysteinylglycyl-γ-methyl-L-glutamyl-N~G-nitro-L-arginylglycyl-L-phenylalanyl-L-phenylalanyl-L-tyrosyl-L-th.reonyl-L-prolyl-s-tosyl-L-lysyl-L-alanine methyl ester(B_(17-30)), a derivative of the C-terminal tetradecapeptide of the B-chain of insulin, has been synthesized either by coupling p-nitrophenyl-carbobenzoxy-L-leucyl-L-valyl-S-benzyl-L-cysteinylglycinate(B_(17-20) ONP) with γ-methyl-L-glutamyl-N~G-nitro-L-arginylglycyl-L-phenylalanyl-L-phenylalanyl-Ltyrosyl-L-threonyl-L-prolyl-e-tosyl-L-lysyl-L-alanine methyl ester or by condensation of carbobenzoxy-L-leucyl-L-valyl-S-benzyl-L-cysteinylglycyl-γ-methyl-L-glutamyl-N~G-nitro-L-arginylglycine (B~(17-23a)) with L-phenylalanyl-L-phenylalanyl-L-tyrosyl-L-threonyl-L-prolyl-e-tosyl-L-lysyl-L-alanine methyl ester(B_(24-30b) by means of the carbodiimide method.The decapeptide derivative, B_(21-30), has been synthesized by condensing carbobenzoxy-γ-methyl-L-glutamyl-N~G-nitro-L-arginylglycine with B_(24-30b) by the use of dicyclohexylcarbodiimide while the heptapeptide derivative, B_(17-23a), has been prepared by the direct coupling of B_(17-20) ONP with γ-methyl-L-glutamyl-N~G-nitroarginylglycine.
