Abstract
The acute and chronic toxicities of N-nitroso-N-methylaniline (NMA) and N-nitroso-N-methylcyclohexylamine (NMC) were compared in rats of both sexes, and LD50 values were determined also for mice and hamsters. NMC is a powerful convulsant in rodents. Chronic administration confirmed both compounds as potent carcinogens acting in the same tissues, with the upper alimentary tract as the main site of action. In view of current biochemical theories for the mechanism of action of nitrosocarcinogens, based on studies with dialkylnitrosamines, the facts that NMA is a strong carcinogen and that NMA and NMC are approximately equipotent, toward the same tissues, are considered significant.
