Abstract
Objective:To a ultra-fast liquid chromatography(UFLC) method for the determination of eugenol and 2-hydroxy-4,6-dimethoxy-5-methylacetophenone in Eugenia caryophyllata.Methods:Shim-pack XR-ODS Ⅱ(3.0 mm×75 mm,2.2 μm) column was adopted with methanol-0.5% phosphoric acid water solution(60∶40) as the mobile phase at a flow rate of 0.8 mL·min-1;The detection wavelength was 280 nm and column temperature was 50 ℃.Results:Both eugenol and 2-hydroxy-4,6-dimethoxy-5-methylacetophenone showed a good linear relationship with peak area within the range of 9.98-79.84 μg(r=0.9999) and 17.08-136.64 μg(r=0.9999),respectively;The average recoveries(n=5) were 99.57% and 98.68%.Conclusion:The UFLC method that is used to quantify eugenol and 2-hydroxy-4,6-dimethoxy-5-methylacetophenone is simple,rapid,accurate,and good reproducibility,so it can be used to evaluate the quality of Eugenia caryophyllata;UFLC method can save analysis time and solvent consumption,thus it can be worthy of promotion to quality the traditional Chinese medicine.
